If screen-lock is not working with short-cut (Ctrl+Alt+L) or menu or widget etc. then use command
gnome-screensaver-command --lock
and see the reason why lock is not working. If the reason is screensaver is not running then run it using
gnome-screensaver
and then try the lock command again.
